• Preparation: This antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human Cyclin E/CCNE1 (rh Cyclin E/CCNE1). The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.
• Preparation and Storage: This antibody can be stored at 2-8 degree C for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20 degree C to -80 degree C. Preservative-Free. Avoid repeated freeze-thaw cycles.

IF (Immunofluorescence)

(Immunofluorescence staining of CCNE1 in Hela cells. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with mouse anti-human CCNE1 monoclonal antibody (dilution ratio 1:60) at 4 degree C overnight. Then cells were stained with the Alexa Fluor488-conjugated Goat Anti-mouse IgG secondary antibody (green) and counterstained with DAPI (blue).Positive staining was localized to nucleolus.)

FCM/FACS (Flow Cytometry)

(Flow cytometric analysis of Human CCNE1 expression on HeLa cells. The cells were treated according to manufacturer's manual (BD Pharmingen'), stained with purified anti-Human CCNE1, then a FITC-conjugated second step antibody. Cyclin E1 is a member of the highly conserved cyclin family and belongs to the E-type cyclin that functions as a regulator of S phase entry and progression in mammalian cells. Cyclin E1 serves as regulatory subunits that bind, activate, and provide substrate for its associated cyclin-dependent kinase2 (CDK2), whose activity is essential for cell cycle G1/S transition. Over expression of this encoding gene has been found in many tumors, which results in chromosome instability and by extension, induce tumorigenesis. This protein was also found to associate with, and be involved in, the phosphorylation of NPAT protein (nuclear protein mapped to the ATM locus), which participates in cell-cycle regulated histone gene expression and plays a critical role in promoting cell-cycle progression in the absence of pRB. In general, cyclin E1, as an activator of phospho-CDK2 (pCDK2), is important for cell cycle progression and is frequently overexpressed in cancer cells.
