Application Notes
Specimen Preparation
Paraffin sections:
The antibody can be used on formalin-fixed paraffin-embedded (FFPE) tissue sections. Ensure tissue undergoes appropriate fixation for best results. Pre-treatment of tissues with heat-induced epitope retrieval (HIER) is recommended using ImmunoDNA Retriever with Citrate, ImmunoDNA Retriever with EDTA or ImmunoDNA Digestor. See reverse side for complete protocol. Tissue should remain hydrated via use of Immuno/DNA Washer solutions.
Frozen sections and cell preparations:
The antibody can be used for labeling acetone-fixed frozen sections and acetone-fixed cell preparations.

Intended Use
This antibody is intended for use in Immunohistochemical applications on formalin fixed paraffin-embedded tissues (FFPE), frozen tissue sections and cell preparations. Interpretation of results should be performed by a qualified medical professional.
Immunogen
Peptide between trans-activation and DNA-binding domains of GATA-3.
Staining Procedure
1. Cut and mount 3-5 micron formalin-fixed paraffin-embedded tissues on positively charged slides such as Hydrophilic Plus Slides.
2. Air dry for 2 hours at 58° C.
3. Deparaffinize, dehydrate and rehydrate tissues.
4. Subjecttissuesto heat induced epitope retrieval (HIER) using a suitable retrieval solution such as ImmunoDNA Retriever with Citrate or EDTA.
5. Any of three heating methods may be used:
a. TintoRetriever Pressure Cooker or Equivalent
Place tissues/slides in a staining dish or coplin jar containing the ImmunoDNA Retriever with Citrate or EDTA, and place on trivet in the pressure cooker. Add 1-2 inches of distilled waterto the pressure cooker and turn heatto high. Incubate for 15 minutes. Open and immediately transfer slides to room temperature.
b. TintoRetriever PT Module or Water Bath Method
Place tissues/slides in a pre-warmed staining dish or coplin jar containing the ImmunoDNA Retriever with Citrate or EDTA at 95°-99° C. Incubate for 30-60 minutes.
c. Conventional Steamer Method
Place tissues/slides in a pre-warmed staining dish or coplin jar containing the ImmunoDNA Retriever with Citrate or EDTA in a steamer, cover and steam for 30-60 minutes.
6. After heattreatment, transfer slides in ImmunoDNA Retriever with Citrate or EDTA to room temperature and let stand for 15-20 minutes.
7. For manual staining, perform antibody incubation atambienttemperature. For automated staining methods, perform antibody incubation according to instrument manufacturer’s instructions.
8. Wash slides with ImmunoDNA washer or DI water.
9. Continue IHC staining protocol. Wash slides between each step with ImmunoDNA washer solution.
2. Air dry for 2 hours at 58° C.
3. Deparaffinize, dehydrate and rehydrate tissues.

Related Product Information for anti-GATA3 antibody
Trans-acting T-cell-specific transcription factor, GATA-3 is, a protein that in humans is encoded by the GATA3 gene. GATA-3 b regulates luminal epithelial cell differentiation in the mammary gland, is an important regulator of T cell development and plays an important role in endothelial cell biology.GATA-3 is one of the three genes mutated in 10% of breast cancers. Nuclear expression of GATA-3 in breast cancer is considered a marker of luminal cancer in ER+ cancer and luminal androgen responsive cancer in ER-/AR+ tumors. It is highly coexpressed with FOXA1 and serves as negative predictor of basal subtype and HER-2 and is also considered a strong predictor of taxane and platin salts insensitivity.GATA3 expression is found in urothelial carcinoma, especially in invasive and high grade tumors. Therefore, anti-GATA3 can be used in a panel of antibodies for identification of unknown primary carcinoma, when carcinomas of the breast or bladder are a possibility. Studies have also shown the utility of GATA-3 in differntiating urothelial carcinoma from prostate adenocarcinoma and squamous cell carcinomas of the uterine, cervix, anus and lung.

Product Notes
The GATA3 gata3 (Catalog #AAA13571) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The GATA3 reacts with Human Predicted: Rat and may cross-react with other species as described in the data sheet. AAA Biotech's GATA3 can be used in a range of immunoassay formats including, but not limited to, Immunohistochemistry (IHC) Frozen/Paraffin. Specimen Preparation Paraffin sections: The antibody can be used on formalin-fixed paraffin-embedded (FFPE) tissue sections. Frozen sections and cell preparations: The antibody can be used for labeling acetone-fixed frozen sections and acetone-fixed cell preparations. Researchers should empirically determine the suitability of the GATA3 gata3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GATA3, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
