WB (Western Blot)

(Western blot analysis of WT1 expression in K562 cell lysate (AAA46902).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-WT1 monoclonal antibody overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for WT1)

Related Product Information for anti-WT1 antibody
Transcription factor that plays an important role in cellular development and cell survival. Regulates the expression of numerous target genes, including EPO. Plays an essential role for development of the urogenital system. Recognizes and binds to the DNA sequence 5'-CGCCCCCGC-3'. It has a tumor suppressor as well as an oncogenic role in tumor formation. Function may be isoform-specific: isoforms lacking the KTS motif may act as transcription factors. Isoforms containing the KTS motif may bind mRNA and play a role in mRNA metabolism or splicing. Isoform 1 has lower affinity for DNA, and can bind RNA.

Product Notes

The WT1 wt1 (Catalog #AAA46902) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Wilms Tumor Protein Rabbit Monoclonal Antibody reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Wilms Tumor Protein can be used in a range of immunoassay formats including, but not limited to, WB (Western Blot), ICC (Immunocytochemistry), IHC (Immunohistochemistry), IF (Immunofluorescence), FCM/FACS (Flow Cytometry). Researchers should empirically determine the suitability of the WT1 wt1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Wilms Tumor Protein, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
