Principle of the Assay: The LumiAb Murine sRANK Ligand Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Murine sRANK Ligand concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Murine sRANK Ligand while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Murine sRANK-Ligand or Soluble Receptor Activator of NF-kappaB Ligand, also known as Tumor Necrosis Factor Ligand Superfamily Member 11, is a 316 amino acid cytokine protein expressed from the Tnfsf11 gene located on chromosome 14. The peptide chain that exists from residue 1-36 is the membrane form of the RANK-Ligand while the chain that exists from residue 139-178 is the soluble form of the RANK-Ligand. The soluble form of isoform 1 derives from the membrane form by proteolytic processing. The cleavage may be catalyzed by ADAM17. The sRANK-Ligand is the cytokine that binds to TNFRSF11B/OPG and to TNFRSF11A/RANK and acts as an osteoclast differentiation and activation factor. Moreover, the sRANK-Ligand homotrimer augments the ability of dendritic cells to stimulate naive T-cell proliferation, and thus may be an important regulator of interactions between T-cells and dendritic cells as well as T-cell-dependent immune responses. It is highly expressed in thymus and lymph nodes, but not in non-lymphoid tissues and is abundantly expressed in T-cells but not in B-cells; a high level expression is also seen in the trabecular bone and lung. Deficiency in Tnfsf11 results in failure to form lobulo-alveolar mammary structures during pregnancy, resulting in death of newborns. Trance-deficient mice show severe osteopetrosis, with no osteoclasts, marrow spaces, or tooth eruption, and exhibit profound growth retardation at several skeletal sites, including the limbs, skull, and vertebrae and have marked chondrodysplasia, with thick, irregular growth plates and a relative increase in hypertrophic chondrocytes.
