Background: DLL1(DELTA-LIKE 1), also known as DL1 or DELTA1, is a protein that in humans is encoded by the DLL1 gene. DLL1 is a human homolog of the Notch Delta ligand and is a member of the delta/serrate/jagged family. It plays a role in mediating cell fate decisions during hematopoiesis, and it is also important in cell-to-cell communication. This gene is mapped to chromosome 6q27, near a locus for type I diabetes. Functional analysis suggested that a soluble fusion protein containing the DSL domain of DLL1 and its adjacent 50 N-terminal amino acids increased the viability of hemopoietic cells but inhibited cell death. It has been found that DLL1 blocks the differentiation of progenitor cells into the B-cell lineage while promoting the emergence of a population of cells with the characteristics of a T-cell/NK-cell precursor.
Principle of the Assay: The Picokine™ Mouse Dll1 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Mouse Dll1 with a 96-well strip plate that is pre-coated with antibody specific for Dll1. The detection antibody is a biotinylated antibody specific for Dll1. The capture antibody is a monoclonal antibody from rat and the detection antibody is a biotinylated polyclonal antibody from goat. The kit includes Mouse Dll1 protein as standards. To measure Mouse Dll1, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Mouse Dll1 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Dll1 in the sample.
