Background: Factor XII (aka Hageman Fact or) is a single-chain, 615 amino acid glycoprotein zymogen [1). Factor XII is activated by kallikrein [2). Factor Xlla converts prekall ikrein to kallikrein during the intrinsic pathway of the coagulation cascade. Although Fact or XII is not thought 10 play en essential role in norm al hemostasis, lack of Factor XII in a mouse model resulted in a " evere defect' in thrombus formation [3].
Principle of the Assay: Mouse Factor XII will bind to the affinity purified capture antibody coated on the microtiter plate. Factor XII and Xlla will react with the antibody on the plate. After appropriate washing steps, biotin labeled anti-mouse Factor XII primary antibody binds to the captured protein. Excess primary antibody is washed away and bound antibody is reacted with peroxidase conjugated streptavidin. Following an additional washing step, TMB substrate is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of mouse Factor XII. Color development is proportional to the concentration of Factor XII in the samples.
