Principle of the Assay: IFN-gamma is the prototype proinflammatory cytokine and is produced by a variety of immune cells under inflammatory conditions,notably by T cells and NK cells. It plays a key role in host defense by promoting the development and activation of Th1 cells,chemoattraction and activation of monocytes and macrophages.Mouse IFN-gamma ELISA Kit employs a double antibodysandwich method to quantitate Mouse IFN-gamma in samples. An antibody specific for Mouse IFN-gamma has been pre-coated onto amicroplate. Standards and samples are pipetted into the wells and any Mouse IFN-gamma present is bound by the immobilizedantibody. After removing any unbound substances, a biotin-conjugated antibody specific for Mouse IFN-gamma is added to the wells.After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unboundstreptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow afteradding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The IFN-gamma concentration isproportional to the OD450 nm value.
