Principle of the Assay: Interleukin 4 is a pleiotropic cytokine produced primarily by activated T lymphocytes, mast cells and basophils.IL-4 has multipleimmune response-modulating functions on a variety of cell types. It is an important modulator of the differentiation of precursor Thelper cells to the Th2 subset that mediates humoral immunity and modulates antibody production.Mouse IL-4 ELISAKit employs a double antibody sandwich method to quantitate Mouse IL-4 in samples. An antibody specific for Mouse IL-4 hasbeen pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Mouse IL-4 present is bound bythe immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Mouse IL-4 is addedto the wells. After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove anyunbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellowafter adding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The IL-4 concentration isproportional to the OD450 nm value.
