Background: Nidogen-1 (NID-1), also known as Entactin, is a protein that in humans is encoded by the NID1 gene. It is a member of the nidogen family of basement membrane glycoproteins. This gene is mapped to 1q42.3. Nidogen-1 is a component of the basement membrane alongside other components such as collagen type IV, proteoglycans (heparan sulfate and glycosaminoglycans), laminin and fibronectin. The protein interacts with several other components of basement membranes. Structurally it (along with perlecan) connects the networks formed by collagens and laminins to each other. It may also play a role in cell interactions with the extracellular matrix. Nidogen-1 also can serve as a bridge between the 2 most abundant molecules in the basement membrane: type IV collagen and laminin.

Principle of the Assay: The Human Nidogen 1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Nidogen 1 in Cell Culture Supernatants, Serum. This assay employs an antibody specific for Human Nidogen 1 coated on a 96-well plate. Standards and samples are pipetted into the wells and Nidogen 1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Nidogen 1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Nidogen 1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Product Notes

The Human NID1 nid1 (Catalog #AAA17881) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA17881 ELISA Kit recognizes Human NID1. AAA Biotech's Nidogen 1 can be used in a range of immunoassay formats including, but not limited to, Sandwich ELISA (SE). Researchers should empirically determine the suitability of the NID1 nid1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Nidogen 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
