Principle of the Assay: The AAA Biotech' Human Nesfatin-1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Nesfatin-1 in Cell Culture Supernatants, Serum. This assay employs an antibody specific for Human Nesfatin-1 coated on a 96-well plate. Standards and samples are pipetted into the wells and Nesfatin-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Nesfatin-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Nesfatin-1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Nesfatin-1 is a metabolic polypeptide encoded in the N-terminal region of the protein precursor, Nucleobindin2 (NUCB2). Recombinant human Nesfatin-1 is a 9.7 kDa protein containing 82 amino acid residues. Originally identified as a hypothalamic neuropeptide, Nesfatin-1 is also expressed in other areas of the brain, and in pancreatic islets of Langerhans, gastric endocrine cells and adipocytes. Nesfatin-1 plays a role in hunger and energy regulation in an independent manner.
