WB: The detection limit for beta Amyloid is approximately 0.25ng/lane reducing conditions.
Tested Species: In-house tested species with positive results.
Predicted Species: Species predicted to be fit for the product based on sequence similarities.
By Heat: Boiling the paraffin sections in 10mM citrate buffer , pH 6.0 , for 20mins is required for that staining of formalin/paraffin sections.
Other applications have not been tested.
Optimal dilutions should be determined by end users.
Immunogen
A synthetic peptide corresponding to a sequence at the C-terminus of human beta Amyloid (672-713aaDAEFRHDSGYEVHHQKLVFFAEDVGSNKGAIIGLMVGGVVIA), different from the related mouse and rat sequences by three amino acids.
Contents
Each vial contains 5mg BSA, 0.9mg NaCl, 0.2mg Na2HPO4, 0.05mg NaN3.
Reconstitution
Add 0.2ml of distilled water will yield a concentration of 500ug/ml.
Relevant Detection Systems
The lab recommends Enhanced Chemiluminescent Kit with anti-Rabbit IgG ( ) for Western blot, and HRP Conjugated anti-Rabbit IgG Super Vision Assay Kit ) for IHC(P), IHC(F) and ICC.
Preparation and Storage
At -20 degree C for one year. After reconstitution, at 4 degree C for one month. It can also be aliquotted and stored frozen at -20 degree C for a longer time. Avoid repeated freezing and thawing.

WB (Western Blot)

The sample well of each lane was loaded with 50ug of sample under reducing conditions. lane 1: rat brain tissue lysate, lane 2: mouse brain tissue lysate. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-beta Amyloid antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for beta Amyloid at approximately 87KD. The expected band size for beta Amyloid is at 87KD.)

Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. Optimal dilutions should be determined by end users. Researchers should empirically determine the suitability of the APP app for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "beta Amyloid, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
