Western blot analysis of CD98/Slc3a2 using anti-CD98/Slc3a2 antibody (AAA126176).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: rat pancreas tissue lysates,Lane 2: rat kidney tissue lysates,Lane 3: mouse pancreas tissue lysates,Lane 4: mouse kidney tissue lysates,Lane 5: mouse testis tissue lysates,Lane 6: mouse NIH/3T3 whole cell lysates.After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD98/Slc3a2 antigen affinity purified polyclonal antibody (#AAA126176) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CD98/Slc3a2 at approximately 80-90 kDa. The expected band size for CD98/Slc3a2 is at 68 kDa.)

This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The kit contains recombinant Mouse Fcgr2b/Fcgr3 with immunogen: Expression system for standard: NS0; Immunogen sequence: T40-R217. To measure Mouse Fcgr2b/Fcgr3, add standards and samples to the wells, then add antibody cocktail. Wash the wells with PBS or TBS buffer, and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product is linearly proportional to Mouse Fcgr2b/Fcgr3 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Fcgr2b/Fcgr3 in the sample.
