Primary Antibody:Rabbit Anti- Connexin-40 antibody AAA268983 , Dilution: 1ug in 100 uL 1X PBS containing 0.5% BSA; Isotype Control Antibody: Rabbit IgG(orange) ,used under the same conditions ); Secondary Antibody: Goat anti-rabbit IgG-PE(white blue), Dilution: 1:200 in 1 X PBS containing 0.5% BSA. Protocol: The cells were fixed with 2% paraformaldehyde (10 min). Antibody were incubated for 30 min on the ice, followed by 1 X PBS containing 0.5% BSA + 1 0% goat serum (15 min) to block non-specific protein-protein interactions. Then the Goat Anti-rabbit IgG/PE antibody was added into the blocking buffer mentioned above to react with the primary antibody of ER1906-57 at 1/200 dilution for 30 min on ice. Acquisition of 20,000 events was performed.)

This gene is a member of the connnexin, gene family. The encoded protein is a component of gap junctions which are composed of arrays of intercellular channels that provide a route for the difussion of low molecular weight materials from cell to cell. Mutations in this gene may be associated with atrial fibrillation. Alternatively spliced transcript variant encoding the same isoform have been described. One gap junction consists of a cluster of closely packed pairs of transmembrane channels, the connnexons, through which materials of low MW diffuse from one cell to a neighboring cell.
