The membrane was incubated with rabbit anti-hnRNP K/HNRNPK antigen affinity purified polyclonal antibody (#AAA126175) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for hnRNP K/HNRNPK at approximately 60 kDa. The expected band size for hnRNP K/HNRNPK is at 55 kDa.)

Background: Chemokine(C-C motif) ligand 7(CCL7) is a small cytokine known as a chemokine that was previously called monocyte-specific chemokine 3(MCP3). It belongs to the C-C chemokine family. By fluorescence in situ hybridization, mapped the MCP3 gene to chromosome 17q11.2-q12. MCP3 was identified as a physiologic substrate of gelatinase A. Cleaved MCP3 binds to CC-chemokine receptors-1, -2, and -3, but no longer induces calcium fluxes or promotes chemotaxis, and instead acts as a general chemokine antagonist that dampens inflammation, suggested that matrix metalloproteinases are both effectors and regulators of the inflammatory response.

Principle of the Assay: The Quick Picokine™ Rat CCL7 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Rat CCL7 in cell culture supernatants, cell lysates, serum and plasma (heparin, EDTA). It uses our proprietary Quick ELISA technology. Quick ELISA technology employs capture antibodies conjugated to an affinity tag that is recognized by the polyclonal antibody used to coat our Quick ELISA plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. The kit contains recombinant Rat CCL7 with immunogen: Expression system for standard: E.coli; Immunogen sequence: Q24-P97. To measure Rat CCL7, add standards and samples to the wells, then add antibody cocktail. Wash the wells with PBS or TBS buffer, and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product is linearly proportional to Rat CCL7 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Rat CCL7 in the sample.

Product Notes

The HNRNPK hnrnpk (Catalog #AAA126175) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-hnRNP K/HNRNPK Antibody Picoband reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's hnRNP K/HNRNPK can be used in a range of immunoassay formats including, but not limited to, ELISA, FCM/FACS (Flow Cytometry), IF (Immunofluorescence), ICC (Immunocytochemistry), WB (Western Blot). Researchers should empirically determine the suitability of the HNRNPK hnrnpk for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "hnRNP K/HNRNPK, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
