Antibodies: Affinity purified rabbit anti-MSK1 antibody AAA211996 used for WB at 0.04 ug/ml (A) and 1 ug/ml (B) and used for IP at 3 ug/mg lysate. MSK1 was also immunoprecipitated by rabbit anti-MSK1 antibody BL9980, which recognizes an upstream epitope. Detection: Chemiluminescence with exposure times of 3 minutes (A) and 10 seconds (B).)

MSK1 is a serine/threonine kinase required for the mitogen or stress-induced phosphorylation of the transcription factors CREB (cAMP response element-binding protein) and ATF1 (activating transcription factor-1). MSK1 Directly represses transcription via phosphorylation of 'Ser-1' of histone H2A. MSK1 also phosphorylates 'Ser-10' of histone H3 in response to mitogenics, stress stimuli and epidemal growth-factor (EGF), which results in the transcriptional activation of several immediate early genes, including proto-oncogenes c-fos/FOS and c-jun/JUN.
