The PCK1 gene (Pck1 in rodents) encodes the cytosolic isozyme of phosphoenolpyruvate carboxykinase (PEPCK-C), which is well-known for its function as a gluconeogenic enzyme in the liver and kidney. PCK1 is a key gene in gluconeogenesis and glyceroneogenesis, and gluconeogenesis mainly occurs in the liver during fasting. Previous studies have demonstrated that PCK1 is downregulated in hepatocellular carcinoma (HCC) and predicts poor prognosis. Overexpression of PCK1 has been shown to suppress liver tumor growth. PCK1 is a key gene in maintaining plasma glucose homeostasis during fasting and that the upregulated expression of PCK1 may be responsible for constant plasma free fatty acid level by the glyceroneogenesis process. PCK1 is a multi-functional enzyme that plays important role in physiological processes, including reproduction.
