IF (Immunofluorescence)

(Fluorescent confocal image of SY5Y cells stained with phospho-Sox2- S246 antibody. SY5Y cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with AAA286283 phospho-Sox2- S246 primary antibody (1:200, 2 h at room temperature). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 ug/ml, 5 min). Note the highly specific localization of the phospho-Sox2 immunosignal mainly to the nucleus.)

This intronless gene encodes a member of the SRY-related
HMG-box (SOX) family of transcription factors involved in the
regulation of embryonic development and in the determination of
cell fate. The product of this gene is required for stem-cell
maintenance in the central nervous system, and also regulates gene
expression in the stomach. Mutations in this gene have been
associated with optic nerve hypoplasia and with syndromic
microphthalmia, a severe form of structural eye malformation. This
gene lies within an intron of another gene called SOX2 overlapping
transcript (SOX2OT).
