Background: The lipid kinase PIK3C3 (also called Vps34) is a class III phosphatidylinositol-3-kinase that specifically catalyzes the formation of phosphatidylinositol-3-phosphate (PI3P) and plays important roles in intracellular membrane trafficking and autophagy. Autophagy is a tightly regulated intracellular process in which portions of cytoplasm, including proteins and organelles, are sequestered within double-membrane vesicles termed autophagosomes and delivered to lysosomes for degradation and recycling. Mammalian cells are postulated to utilize autophagy as a mechanism for turnover of long-lived proteins, removal of protein aggregates and damaged organelles, and as a survival strategy under metabolic stress conditions such as starvation. Among the autophagy machineries, PIK3C3/VPS34 complex functions as an autophagy-triggering kinase to recruit the subsequent autophagy protein machineries on the phagophore membrane. PIK3C3/VPS34 (phosphatidylinositol 3-kinase catalytic subunit type 3) converts phosphatidylinositol (PtdIns) to phosphatidylinositol-3-phosphate (PtdIns3P), sustaining macroautophagy/autophagy and endosomal transport. The PIK3C3/VPS34 subunit of the class III phosphatidylinositol 3-kinase (PtdIns3K) complex is a key early player in macroautophagy/autophagy.
