WB (Western Blot)

(Detection of human SCAP by western blot. Samples: Whole cell lysate (10 ug) from HEK293T, Hep-G2, HeLa, Jurkat, and K-562 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-SCAP antibody (AAA212222 lot 2) used for WB at 0.04 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.)

IP (Immunoprecipitation)

(Detection of human SCAP by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HEK293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-SCAP antibody (AAA212222 lot 2) used for IP at 6 ug per reaction. SCAP was also immunoprecipitated by a previous lot of this antibody (AAA212222 lot 1). For blotting immunoprecipitated SCAP, AAA212222 was used at 0.1 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.)

Sterol regulatory element-binding protein cleavage-activating protein (SCAP) is a protein with a sterol sensing domain (SSD) and seven WD domains. In the presence of cholesterol, SCAP binds to sterol regulatory element binding proteins (SREBPs) and mediates their transport from the ER to the Golgi. The SREBPs are then proteolytically cleaved and regulate sterol biosynthesis [taken from NCBI Entrez Gene (Gene ID: 22937)].
