• Function: Subunit of the V0 complex of vacuolar(H+)-ATPase (V-ATPase), a multisubunit enzyme composed of a peripheral complex (V1) that hydrolyzes ATP and a membrane integral complex (V0) that translocates protons (By similarity). V-ATPase is responsible for acidifying and maintaining the pH of intracellular compartments and in some cell types, is targeted to the plasma membrane, where it is responsible for acidifying the extracellular environment (By similarity). Seems to be directly involved in T-cell activation (PubMed:10329006).

FCM/FACS (Flow Cytometry)

(Overlay histogram showing U-2 OS cells stained with AAA290895 (green line). The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were then icubated in 2% bovine serum albumin to block non-specific protein-protein interactions followed by the antibody (AAA290895, 1:25 dilution) for 60 min at 37 degree C. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight 488 Conjugated Highly Cross-Adsorbed(OE188374) at 1/200 dilution for 40 min at 37 degree C. Isotype control antibody (blue line) was rabbit IgG1 (1mug/1x10^6 cells) used under the same conditions. Acquisition of >10, 000 events was performed.)
