Specificity
The FAK (Phospho-Tyr397) Phospho Sandwich ELISA Kit is a convenient, lysate- free, high throughput and sensitive assay kit that can detect FAK protein phosphorylation and expression profile in cell lysates. The kit can be used for measuring the relative amounts of phosphorylated FAK in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on FAK phosphorylation.

Principle of the Assay: The Phospho Sandwich FAK (Phospho-Tyr397) ELISA Kit contains the components necessary for semi-quantitative determination of FAK (Phospho-Tyr397) concentrations within experimental cell lysate samples. This particular immunoassay utilizes the traditional "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope FAK (Phospho-Tyr397) while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a colorimetric reaction to ensue upon substrate addition. When the substrate TMB (3, 3', 5, 5'-Tetramethylbenzidine) is added, the reaction catalyzed by peroxidase yields a blue color that is representative of the antigen concentration. Upon sufficient color development, the reaction can be terminated through addition of Stop Solution (2 N Sulfuric Acid) where the color of the solution will turn yellow. The absorbance of each well can then be read by a spectrophotometer where data analysis may begin.

Product Notes
The Human, Mouse, Rat PTK2 ptk2 (Catalog #AAA318617) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA318617 ELISA Kit recognizes Human, Mouse, Rat PTK2. It is sometimes possible for the material contained within the vial of "FAK, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
