Principle of the Assay: The AAA Biotech's Mouse RSPO3 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse RSPO3 in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Mouse RSPO3 coated on a 96-well plate. Standards and samples are pipetted into the wells and RSPO3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse RSPO3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of RSPO3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm!!Background/Introduction: R-spondin-3 is a protein that in humans is encoded by the RSPO3 gene. This gene belongs to the R-spondin family. The encoded protein plays a role in the regulation of Wnt (wingless-type MMTV integration site family)/beta-catenin and Wnt/planar cell polarity (PCP) signaling pathways, which are involved in development, cell growth and disease pathogenesis. Genome-wide association studies suggest a correlation of this gene with bone mineral density and risk of fracture. This gene may be involved in tumor development.
