Background: Secreted phosphoprotein 24 (spp24) is a member of the cystatin superfamily, which was first identified in cattle as a minor component of cortical bone and subsequently has been identified as a component of the fetuin-mineral complex.
Principle of the Assay: Rat Secreted Phosphoprotein 24 (SPP2) ELISA Kit employs a two-site sandwich ELISA to quantitate SPP2 in samples. An antibody specific for SPP2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SPP2 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Rat SPP2 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of SPP2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
