Short Description: The peptide corresponding to amino acids 1522-1656 that contains the bromodomain sequences of TAF1 (accession number NP_004597.2) was expressed in E Coli and contains an N-terminal His-tag and C-terminal DYKDDDDK tag with an observed molecular weight of 21.75 kDa. It shows binding specificity for acetylated H3K14. TAF1 RNA polymerase II, TATA box binding protein (TBP)-associated factor (TAF1) is the largest component and core scaffold of the TFIID basal transcription factor complex. TAF1 contains novel N- and C-terminal Ser/Thr kinase domains which can autophosphorylate or transphosphorylate other transcription factors. TAF1 also contains bromodomains for interaction with other proteins. The bromodomain functions as a 'reader' of epigenetic histone marks and regulates chromatin structure and gene expression by linking associated proteins to the recognized acetylated nucleosomal targets.
