Background: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of GSDMD in human serum, plasma, cell culture supernatants and other biological fluids.
Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. An antibody specific for human GSDMD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GSDMD present is bound by the immobilized antibody. After washing away any unbound substances, and then a detection antibody specific for GSDMD is added to the wells and binds to the combination of capture antibody GSDMD in sample. Following a wash to remove any unbound combination, and enzyme conjugate is added to the wells. Following incubation and wash steps, a substrate solution is added to the wells and color develops in proportion to the amount of GSDMD bound in the initial step. The color development is stopped and the absorbance is measured.
