Principle of the Assay: The Rat E Cadherin ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat E Cadherin in Cell Culture Supernates, Serum, Plasma. This assay employs an antibody specific for Rat E Cadherin coated on a 96-well plate. Standards and samples are pipetted into the wells and E Cadherin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat E Cadherin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of E Cadherin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Downregulation of E-cadherin expression often correlates with strong invasive potential and poor prognosis of human carcinomas. The gene spans a region of approximately 100 kb, and its location on chromosome 16q22.1. It contains 16 exons and a 65-kb-long intron 2. E-cadherin gene mutations may contribute to the development of diffusely growing gastric carcinomas. It plays a central part in the process of epithelial morphogenesis and acts as a strong invasion suppressor in experimental tumor cell systems. The standard product used in this kit is recombinant gene expression with the molecular mass of 120KDa.
