Background: Mammalian mitochondrial ribosomal proteins are encoded by nuclear genes and help inprotein synthesis within the mitochondrion. Mitochondrial ribosomes (mitoribosomes) consist of a small 28S subunit and a large 39S subunit. They have an estimated 75%protein to rRNA composition compared to prokaryotic ribosomes, where this ratio is reversed. Another difference between mammalian mitoribosomes and prokaryotic ribosomes is that the latter contain a 5S rRNA. Among different species, the proteins comprising the mitoribosome differ greatly in sequence, and sometimes in biochemical properties, which prevents easy recognition by sequence homology.
Principle of the Assay: Human 28S ribosomal protein S23, mitochondrial (MRPS23) ELISA Kit employs a two-sitesandwich ELISA to quantitate MRPS23 in samples. An antibody specific for MRPS23 hasbeen pre-coated onto a microplate. Standards and samples are pipetted into the wells andany MRPS23 present is bound by the immobilized antibody. After removing any unboundsubstances, HRP-Conjugated Human MRPS23 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is addedto the wells and color develops in proportion to the amount of MRPS23 bound in the initial step. The color development is stopped and the intensity of the color is measured.
