Principle of the Assay: CCL2 gene is one of several cytokine genes clustered on the q-arm of chromosome 17. Cytokines are a family of secretedproteins involved in immunoregulatory and inflammatory processes. The protein encoded by this gene is structurally related to theCXC subfamily of cytokines.Human CCL2 ELISA Kit employs a double antibody sandwich method to quantitate HumanCCL2 in samples. An antibody specific for Human CCL2 has been pre-coated onto a microplate. Standards and samples arepipetted into the wells and any Human CCL2 present is bound by the immobilized antibody. After removing any unboundsubstances, a biotin-conjugated antibody specific for Human CCL2 is added to the wells. After washing, proprietaryStreptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent,adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measurethe OD value with a microplate reader at 450 nm wavelength. The Human CCL2 concentration is proportional to the OD450 nmvalue.
