Background: Chemokine (C-X-C motif) ligand 10 (CXCL10) or IP-10 is a small cytokine belonging to the CXC chemokine family that is also known as 10 kDa interferon-gamma-induced protein (gamma-IP10 or IP-10). CXCL10 is secreted by several cell types in response to IFN-gamma. These cell types include monocytes, endothelial cells and fibroblasts. CXCL10 has been attributed to several roles, such as chemoattraction for monocytes/macrophages, T cells, NK cells, and dendritic cells, promotion of T cell adhesion to endothelial cells, antitumor activity, and inhibition of bone marrow colony formation and angiogenesis.It is a potent inhibitor of angiogenesis in vivo. The gene for CXCL10 is located on human chromosome 4 in a cluster among several other CXC chemokines. It is a RAS target gene and is overexpressed in the majority of colorectal cancers.This chemokine elicits its effects by binding to the cell surface chemokine receptor CXCR3.

Principle of the Assay: The Picokine™ Human CXCL10 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Human CXCL10 with a 96-well strip plate that is pre-coated with antibody specific for CXCL10. The detection antibody is a biotinylated antibody specific for CXCL10. The capture antibody is monoclonal antibody from mouse and the detection antibody is polyclonal antibody from goat. The kit contains recombinant Human CXCL10 with immunogen: Expression system for standard: E.coli; Immunogen sequence: V22-P98. The kit is analytically validated with ready-to-use reagents. To measure Human CXCL10, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Human CXCL10 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human CXCL10 in the sample.
