Principle of the Assay: Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis.Human MMP-9 ELISA Kit employs a double antibody sandwich method to quantitate Human MMP-9 in samples. An antibody specific for Human MMP-9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Human MMP-9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Human MMP-9 is added to the wells. After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent, add ing HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The MMP-9 con centrat ion is proporti onal to the OD450 nm value.
