Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.

Description: This Human Tumor necrosis factor alpha-induced protein 3 (TNFAIP3) ELISA Kit employs a two-site sandwich ELISA to quantitate TNFAIP3 in samples. An antibody specific for TNFAIP3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TNFAIP3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TNFAIP3 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TNFAIP3 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background: Tumor necrosis factor, alpha-induced protein 3 is a zinc finger protein, and has been shown to inhibit NF-kappa B activation as well as TNF-mediated apoptosis. Knockout studies of a similar gene in mice suggested that this gene is critical for limiting inflammation by terminating TNF-induced NF-kappa B responses. Essential component of a ubiquitin-editing protein complex, comprising also RNF11, ITCH and TAX1BP1, that ensures the transient nature of inflammatory signaling pathways. Upon TNF stimulation, deubiquitinates 'Lys-63'-polyubiquitin chains on RIPK1 and catalyzes the formation of 'Lys-48'-polyubiquitin chains. This leads to RIPK1 proteosomal degradation and consequently termination of the TNF- or LPS-mediated activation of NF-kappa-B.

Product Notes
The Human TNFAIP3 tnfaip3 (Catalog #AAA330453) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA330453 ELISA Kit recognizes Human TNFAIP3. It is sometimes possible for the material contained within the vial of "Tumor Necrosis Factor alpha-Induced Protein 3 (TNFAIP3), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
