Recombinant JMJD3/KDM6B (1043-1682) is suitable for use in the study of enzyme kinetics, inhibitor screening, and selectivity profiling. HDM Activity Assay Conditions: 1uM H3K27me3 peptide was incubated with different concentrations of JMJD3 (1043- 1682) protein in 10ul reaction system containing 50mM HEPES-NaOH pH 7.4, 0.02% Triton X-100, 1mM TCEP, 50uM 2-OG, 50uM Ascorbate and 25uM (NH4)2Fe(SO4)2·6H2O for 2 hours, then 10 micro;l H3K27me2 antibody and SA-XL665 mixture (each 1:100 dilution in HTRF Detection Buffer) was added to each reaction system and incubated for 30 min. All the operations and reactions were performed at room temperature. HTRF assay was used for detection.
