Principle of the Assay: IL-6 is a multifunctional protein produced by lymphoid and non-lymphoid cells, and by normal and transformed cells, including Tcells, monocyte/macrophages, fibroblasts, hepatocytes, vascular endothelial cells, cardiac myxomas, bladder cell carcinomas,myelomas, astrogliomas and glioblastomas.Mouse IL-6 ELISA Kit employs a double antibody sandwich method toquantitate Mouse IL-6 in samples. An antibody specific for Mouse IL-6 has been pre-coated onto a microplate. Standards andsamples are pipetted into the wells and any Mouse IL-6 present is bound by the immobilized antibody. After removing anyunbound substances, a biotin-conjugated antibody specific for Mouse IL-6 is added to the wells. After washing, proprietaryStreptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzyme reagent,adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution. Measurethe OD value with a microplate reader at 450nm wavelength. The IL-6 concentration is proportional to the OD450 nm value.
