Principle of the Assay: TGF-beta is capable of producing a variety of effects and virtually all cell types respond to this factor in some way. The inappropriatepresence of active TGF-beta1 has been implicated in a variety of pathological conditions Because of the necessity for regulating itsactivity tightly, TGF-beta is secreted by cells in the form of an inactive complex.Mouse TGF-beta1 ELISA Kit employs adouble antibody sandwich method to quantitate Mouse TGF-beta1 in samples. An antibody specific for Mouse TGF-beta1 has beenpre-coated onto a microplate. Standards and samples are pipetted into the wells and any Mouse TGF-beta1 present is bound by theimmobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Mouse TGF-beta1 is addedto the wells. After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove anyunbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellowafter adding stop solution. Measure the OD value with a microplate reader at 450nm wavelength. The TGF-beta1 concentration isproportional to the OD450 nm value.
