Post-Translational Modifications
ATM is phosphorylated by NUAK1/ARK5. Autophosphorylation occurs on Ser-367, Ser-1893, Ser-1981 and correlates with DNA damage-mediated activation of the kinase. Acetylation, on DNA damage, is required for activation of the kinase activity, dimer-monomer transition, and subsequent autophosphorylation on Ser-198. ATM is acetylated in vitro by KAT5/TIP601. Ubiquitination occurs at Lys215, Lys1109, Lys1114, Lys1126, Lys1323, Lys1572, Lys1994, Lys2025, Lys2148, Lys2160, Lys2789 and Lys2848.

Biological Significance: ATM is a serine/threonine protein kinase that is recruited and activated by DNA double-strand breaks. It phosphorylates several key proteins that initiate activation of the DNA damage checkpoint, leading to cell cycle arrest, DNA repair or apoptosis. Several of these targets, including p53, CHK2 and H2AX are tumor suppressors.
Sequence Info: ATM is composed of 3056 amino acids with a molecular weight of 350687 Da. There is a FAT domain at amino acids 1960 2566, a PI3K/PI4K domain at amino acids 2712 2962 and a FATC domain at amino acids 3024 3056.
Implications in Disease: Defects in ATM expression have been closely linked with Ataxia telangiectasia (AT), a rare human disease characterized by cerebellar degeneration, extreme cellular sensitivity to radiation and a predisposition to cancer. All AT patients contain mutations in the ATM gene. Defects in ATM result in severe consequences in repairing certain types of damage to DNA, and cancer may result from improper repair. AT patients have an increased risk for breast cancer that has been ascribed to ATM's interaction and phosphorylation of BRCA1 and its associated proteins following DNA damage. Certain kinds of leukemias and lymphomas, including Mantle cell lymphoma, T-ALL, atypical B cell chronic lymphocytic leukemia, and T-PLL are also associated with ATM defects.
Expression Note: Expression of defective ATM has been reported in a variety of carcinomas. ATM is found in the pancreas, kidney, skeletal muscle, liver, lung, placenta, brain, heart, spleen, thymus, testis, ovary, small intestine, colon and leukocytes.
