IHC (Immunohiostchemistry)

(Figure 2. IHC analysis of GATA1 using anti-GATA1 antibody (AAA126020).GATA1 was detected in a paraffin-embedded section of human testis tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 ug/ml rabbit anti-GATA1 Antibody (AAA126020) overnight at 4 degree C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37 degree C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen.)

WB (Western Blot)

(Figure 1. Western blot analysis of GATA1 using anti-GATA1 antibody (AAA126020).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: human K562 whole cell lysates,Lane 2: mouse testis tissue lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-GATA1 antigen affinity purified polyclonal antibody (#AAA126020) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for GATA1 at approximately 43-50 kDa. The expected band size for GATA1 is at 43 kDa.)

Related Product Information for anti-GATA1 antibody
GATA-binding factor 1 or GATA-1 (also termed Erythroid transcription factor) is the founding member of the GATA family of transcription factors. The protein plays an important role in erythroid development by regulating the switch of fetal hemoglobin to adult hemoglobin. Mutations in this gene have been associated with X-linked dyserythropoietic anemia and thrombocytopenia.

Product Notes

The GATA1 gata1 (Catalog #AAA126020) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-GATA1 Antibody Picoband reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's GATA1 can be used in a range of immunoassay formats including, but not limited to, WB (Western Blot), IHC (Immunohistochemistry), FCM/FACS (Flow Cytometry), ELISA. Researchers should empirically determine the suitability of the GATA1 gata1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GATA1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
