Preparation and Storage

Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS,blocked with 10% serum, and incubated with rabbit anti-Human NASP polyclonal antibody (dilution ratio 1:200) at 4 degree C overnight. Then cells were stained with the Alexa Fluor488-conjugated Goat Anti-rabbit IgG secondary antibody (green). Positive staining was localized to Nucleus.)

NASP (Nuclear Autoantigenic Sperm Protein) is a linker histone chaperone required for normal cell division. NASP is a histone H1 binding protein expressed in all cells undergoing division. Synchronized HeLa cells are delayed in progression through the G1/S border when transiently transfected to overexpress full-length NASP, but not the histone-binding site (HBS) deletion mutant (NASP-DeltaHBS). Changes in NASP expression significantly affect cell growth and development; loss of gene function results in embryonic lethality. In humans, growing evidence implicates NASP miss-regulation in the development of a variety of cancers. NASP cDNA contains 2561 nt encoding a protein of 787 amino acids. The open reading frame contains 2446 nt followed by an ochre stop codon (TAA) and 104 nucleotides of untranslated sequence containing a poly(A) addition signal 10 bases upstream of the poly(A) tail.
