• Assay Procedure Step 1: Aliquot 0.1ml per well of the 3000pg/ml, 1500pg/ml, 750pg/ml, 375.pg/ml, 187.5pg/ml, 93.8pg/ml, 46.9pg/ml rat DKK1 standard solutions into the precoated 96-well plate. Add 0.1ml of the sample diluent buffer into the control well (Zero well). Add 0.1ml of each properly diluted sample of rat cell culture supernatants, serum or plasma (heparin, EDTA) to each empty well. We recommend that each rat DKK1 standard solution and each sample is measured in duplicate.

Principle of the assay: Rat DKK1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for DKK1 has been precoated onto 96-well plates. Standards(Expression system for standard: NSO, Immunogen sequence: S23-H270) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for DKK1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the rat DKK1 amount of sample captured in plate.

Background: Dickkopf-related protein 1 is a protein that in humans is encoded by the DKK1 gene, which was mapped to chromosome 10q11.2. This gene encodes a protein that is a member of the dickkopf family. It is a secreted protein with two cysteine rich regions and is involved in embryonic development through its inhibition of the WNT signaling pathway. Elevated levels of DKK1 in bone marrow, plasma and peripheral blood is associated with the presence of osteolytic bone lesions in patients with multiple myeloma.
