Principle of the Assay: Interleukin 6 (IL-6) is a multifunctional cytokine that plays important roles in host defense, acute phase reactions, immuneresponses, nerve cell functions and hematopoiesis. It is expressed by a variety of normal and transformed lymphoid andnon-lymphoid cells.Rat IL-6 ELISA Kit employs a double antibody sandwich method to quantitate Rat IL-6 in samples.An antibody specific for Rat IL-6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells andany Rat IL-6 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibodyspecific for Rat IL-6 is added to the wells. After washing, proprietary Streptavidin-HRP conjugates is added to the wells. Followinga wash to remove any unbound streptavidin-enzyme reagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis ofHRP, and turns yellow after adding stop solution. Measure the OD value with a microplate reader at 450 nm wavelength. The RatIL-6 concentration is proportional to the OD450 nm value.
