Background: L-selectin, also known as CD62L, is a cell adhesion molecule found on leukocytes. It belongs to the selectin family of proteins, which recognize sialylated carbohydrate groups. It is cleaved by ADAM17. SELL (L-selectin)is a cell surface component that is a member of a family of adhesion/homing receptors which play important roles in leukocyte-endothelial cell interactions. The molecule is composed of multiple domains: one homologous to lectins, one to epidermal growth factor, and two to the consensus repeat units found in C3/C4 binding proteins. L-selectin acts as a "homing receptor" for leukocytes to enter secondary lymphoid tissues via high endothelial venules. Ligands present on endothelial cells will bind to leukocyte expressing L-selectin, slowing leukocyte trafficking through the blood, and facilitating entry into a secondary lymphoid organ at that point.
Principle of the Assay: The Mouse CD62L ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD62L in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse CD62L coated on a 96-well plate. Standards and samples are pipetted into the wells and CD62L present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD62L antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD62L bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
