Introduction: Receptor-binding cancer antigen expressed on SiSo cells (RCAS1) is a human tumor-associated antigen that induces cell cycle arrest and/or apoptosis in RCAS1 receptor-expressing immune cells. RCAS1 has been identified by means of a monoclonal antibody on the human uterine cervical adenocarcinoma cell line SiSo. RCAS1 is identical with the human estrogen-responsive gene product EBAG-9 (ER-binding fragment-associated antigen-9). RCAS1 is expressed at a high frequency in human uterine and ovarian cancer cells as well as in other mammalian cancer cells. High expression of RCAS1 has been shown to correlate significantly with tumour progression and predicted poor outcome in gallbladder cancer. RCAS1 is detected in nearly 100 % of gastric carcinomas and is strictly associated with tumor progression of gastric cancer. RCAS1 is highly expressed also in extramammary Paget's disease and in advanced stage squamous cell carcinoma of the skin. Ohshima et al have reported that RCAS1 is expressed in Reed-Sternberg cells in association with Epstein-Barr virus infection and that RCAS1 rather than FAS ligand allows these cells to evade host immune responses.
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to RCAS1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for RCAS1 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain RCAS1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of RCAS1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
