Background: Fibrinogen is a soluble glycoprotein t hat circu lates in the blood and is co n ver~ed t o insoluble fibrin by thrombin in t he fina l st ep of the coagu lation cascade [1). Hepatic expression of fibrinogen increases two t o four hundred fo ld during t he acute phase response to infecti on or inflammation [2). Elevated fi brinogen levels are correlated with ca rdiovascular disease [3) and atherosclerosis [4).
Principle of the Assay: Mouse fibrin ogen wi ll bind to t he affinity purified capture antibody coated on the microtiter plat e. After appropriate was hi ng steps, biotin labeled polyclonal anti-mouse fibrinogen primary ant ibody binds to the captured prot ein. Excess antibody is washed away and bound polyclonal antibody is react ed with avidin conjugated to horseradish peroxidase. TMB substrate is used for color development at 4S0nm. A standard ca libration curve is prepared along w ith the samples to be measured using diluti ons of mouse fibrinogen. Color development is proportional t o t he concentration of fibrinogen in th e samples.
