Reconstitution
Centrifuge the vial prior to opening. Reconstitute in water to a concentration of < 0.2 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
Endotoxin level
< 0.1 ng /ug of protein (<1EU/ug)
Biological Activity
Determined by its ability to decrease alkaline phosphatase activity in CCL-226 cells when treated with 25 ng/ml of murine Wnt3a.
Preparation and Storage
The lyophilized protein is stable at room temperature for 1 month and at 4°C for 3 months. Reconstituted working aliquots are stable for 1 week at 2°C to 8°C and for 3 months at -20°C to -80°C.
Related Product Information for Wnt-7a active protein
Wnt-7a belongs to the Wnt family of signaling proteins that play a key role in maintaining the integrity of embryonic and adult tissues. It is expressed in placenta, kidney, testis, uterus, fetal lung and fetal and adult brain. Most Wnt proteins can signal though a mechanism called the canonical Wnt pathway, in which Wnt proteins bind to and activate seven-pass transmembrane receptors of the Frizzled family ultimately leading to the disruption of beta-Catenein degradation. Intracellular accumulation of ï¢-Catenin increases translocation of the protein into the nucleus where it binds to TCF/LEF transcription factors and induces the expression of numerous genes. Increased Wnt/beta-Catenein signaling is associated with tumorigenesis in a diverse set of human cancers. However, Wnt-7a/Frizzled-9 signaling has been shown to act as a tumor suppressor in non-small cell lung cancers. Recombinant human Wnt-7a is a 35.5 kDa glycoprotein containing 318 amino acids. Due to glycosylation, Wnt-7a migrates between 40-55 kDa by SDS-PAGE gel under unreduced conditions

The Human Wnt-7a reacts with Human and may cross-react with other species as described in the data sheet. The amino acid sequence is listed below: LGASIICNKI PGLAPRQRAI CQSRPDAIIV IGEGSQMGLD ECQFQF RNGR WNCSALGERT VFGKELKVGS REAAFTYAII AAGVAHAITA A CTQGNLSDC GCDKEKQGQY HRDEGWKWGG CSADIRYGIG FAKVFVD ARE IKQNARTLMN LHNNEAGRKI LEENMKLECK CHGVSGSCTT KT CWTTLPQF RELGYVLKDK YNEAVHVEPV RASRNKRPTF LKIKKPLS YR KPMDTDLVYI EKSPNYCEED PVTGSVGTQG RACNKTAPQA SGC DLMCCGR GYNTHQYARV WQCNCKFHWC CYVKCNTCSE RTEMYTCK. It is sometimes possible for the material contained within the vial of "Wnt-7a, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
