product-image-AAA71962_WB11.png WB (Western Blot) ( Western Blot using anti-p53 antibody A431 cell nuclear (A) and cytoplasmic (B) extract (35ug protein in RIPA buffer) was resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version at 0.1 ug/ml before detection by an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected band size for p53 is 43.7 kDa, though due to the high number of proline residues in this protein runs at a size of ~53kDa (c.f. Ziemer et al., PMID: 7107651). successfully detected both human nuclear and cytoplasmic p53.)

IF (Immunofluorescence)

( Immunofluorescence staining of fixed A431 with anti-p53 antibody Immunofluorescence analysis of paraformaldehyde fixed A431 cells, permeabilized with 0.15% Triton and stained with the chimeric rabbit IgG version at 10ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (1ug/ml), showing nuclear staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom DAPI, merged channels and a negative control. The negative control was stained with unimmunized rabbit IgG followed by Alexa Fluor 488 secondary antibody.)

Product Notes

The p53 tp53 (Catalog #AAA71962) is a Recombinant Antibody and is intended for research purposes only. The product is available for immediate purchase. The Anti-p53 [PAb421] reacts with Human, Monkey, Mouse, Rabbit, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's p53 can be used in a range of immunoassay formats including, but not limited to, WB (Western Blot), IP (Immunoprecipitation), FCM/FACS (Flow Cytometry), IF (Immunofluorescence), ELISA. Researchers should empirically determine the suitability of the p53 tp53 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "p53, Monoclonal Recombinant Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
