Principle of the Assay: CCL2 is one of several cytokine genes clustered on chromosome 11. Chemokines are a superfamily of secreted proteins involvedin immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement ofN-terminal cysteine residues of the mature peptide.Mouse CCL2 ELISA Kit employs a double antibody sandwichmethod to quantitate Mouse CCL2 in samples. An antibody specific for Mouse CCL2 has been pre-coated onto a microplate.Standards and samples are pipetted into the wells and any Mouse CCL2 present is bound by the immobilized antibody. Afterremoving any unbound substances, a biotin-conjugated antibody specific for Mouse CCL2 is added to the wells. After washing,proprietary Streptavidin-HRP conjugates is added to the wells. Following a wash to remove any unbound streptavidin-enzymereagent, adding HRP Substrate (TMB), TMB turns blue under the catalysis of HRP, and turns yellow after adding stop solution.Measure the OD value with a microplate reader at 450 nm wavelength. The CCL2 concentration is proportional to the OD450 nmvalue.
