WB (Western Blot)

(Figure 1. Western blot analysis of DNA Ligase III/LIG3 using anti-DNA Ligase III/LIG3 antibody (AAA126249).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: human HepG2 whole cell lysates,Lane 2: human HEK293 whole cell lysates,Lane 3: human PC-3 whole cell lysates,Lane 4: human THP-1 whole cell lysates,Lane 5: rat PC-12 whole cell lysates,Lane 6: mouse RAW264.7 whole cell lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-DNA Ligase III/LIG3 antigen affinity purified polyclonal antibody (#AAA126249) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for DNA Ligase III/LIG3 at approximately 113 kDa. The expected band size for DNA Ligase III/LIG3 is at 113 kDa.)

Related Product Information for anti-LIG3 antibody
DNA ligase 3 is an enzyme that, in humans, is encoded by the LIG3 gene. This gene is a member of the DNA ligase family. Each member of this family encodes a protein that catalyzes the joining of DNA ends but they each have a distinct role in DNA metabolism. The protein encoded by this gene is involved in excision repair and is located in both the mitochondria and nucleus, with translation initiation from the upstream start codon allowing for transport to the mitochondria and translation initiation from a downstream start codon allowing for transport to the nucleus. Additionally, alternate transcriptional splice variants, encoding different isoforms, have been characterized.

Product Notes

The LIG3 lig3 (Catalog #AAA126249) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-DNA Ligase III/LIG3 Antibody Picoband reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's DNA Ligase III/LIG3 can be used in a range of immunoassay formats including, but not limited to, WB (Western Blot), FCM/FACS (Flow Cytometry), ELISA. Researchers should empirically determine the suitability of the LIG3 lig3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "DNA Ligase III/LIG3, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
