(Figure 2. IF analysis of LAMTOR1 using anti-LAMTOR1 antibody (AAA126457).LAMTOR1 was detected in an immunocytochemical section of MCF-7 cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent (AR0022) for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 5 ug/mL rabbit anti-LAMTOR1 Antibody (AAA126457) overnight at 4 degree C. DyLight594 Conjugated Goat Anti-Rabbit IgG (BA1142) was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37 degree C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.)

WB (Western Blot)

(Figure 1. Western blot analysis of LAMTOR1 using anti-LAMTOR1 antibody (AAA126457).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: monkey COS-7 whole cell lysates,Lane 2: human MCF-7 whole cell lysates,Lane 3: rat lung tissue lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-LAMTOR1 antigen affinity purified polyclonal antibody (#AAA126457) at 0.25 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for LAMTOR1 at approximately 18 kDa. The expected band size for LAMTOR1 is at 18 kDa.)

Related Product Information for anti-LAMTOR1 antibody
Late endosomal/lysosomal adaptor, MAPK and MTOR activator 1 is a protein that in humans is encoded by the LAMTOR1 gene. mTORC1 kinase complex is a critical component in the regulation of cell growth. Its activity is modulated by energy levels, growth factors, and amino acids. The four related GTPases, RagA, RagB, RagC, and RagD, have been shown to interact with raptor in mTORC1. These interactions are both necessary and sufficient for mTORC1 activation in response to amino acid signals. A protein complex consisting of LAMTOR1/C11orf59, LAMTOR2/ROBLD3, and LAMTOR3/MAPKSP1 has been identified to interact with and recruit the four Rag GTPases to the surface of lysosomes.

Product Notes

The LAMTOR1 lamtor1 (Catalog #AAA126457) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-LAMTOR1 Antibody Picoband reacts with Human, Rat, Monkey and may cross-react with other species as described in the data sheet. AAA Biotech's LAMTOR1 can be used in a range of immunoassay formats including, but not limited to, ELISA, FCM/FACS (Flow Cytometry), IF (Immunofluorescence), ICC (Immunocytochemistry), WB (Western Blot). Researchers should empirically determine the suitability of the LAMTOR1 lamtor1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "LAMTOR1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
