NKX6-1 (NK6 homeobox 1) is a neuronal progenitor transcription factor that is induced by SHH signals. It binds to specific A/T-rich DNA sequences in the promoter regions of a number of genes. It functions in transcriptional regulation in islet beta cells, and binds to the insulin promoter and is involved in regulation of insulin. It restricts the generation of motor neurons to the appropriate region of the neural tube. It is considered a tumor suppressor via its ability to suppress the epithelial-to-mesenchymal transition, and loss of expression is correlated with metastasis in cervical cancer. In immunohistochemistry of normal tissue, NKX6-1 has nuclear positivity in islets of Langerhans in the pancreas, and is also found in the brain, thyroid, stomach, breast and testis.
