WB (Western Blot)

(Figure 1. Western blot analysis of Scavenging Receptor SR-BI/SCARB1 using anti-Scavenging Receptor SR-BI/SCARB1 antibody (AAA126106).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: human T-47D whole cell lysates,Lane 2: mouse liver tissue lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Scavenging Receptor SR-BI/SCARB1 antigen affinity purified polyclonal antibody (#AAA126106) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Scavenging Receptor SR-BI/SCARB1 at approximately 85 kDa. The expected band size for Scavenging Receptor SR-BI/SCARB1 is at 85 kDa.)

Background: Nerve growth factor receptor (NGFR) is also referred to as p75(NTR) because of its ability to bind at low affinity not only to NGF, but also other neurotrophins including brain-derived neurotrophic factor (BDNF), neurotrophin-3 and neurotrophin-4/5. The nerve growth factor receptor gene is at human chromosome region 17q12-17q22, distal to the chromosome 17 breakpoint in acute leukemias. The neurotrophin receptor p75(NTR), a tumor necrosis factor receptor superfamily member expressed in hepatic stellate cells after fibrotic and cirrhotic liver injury in humans, is a regulator of liver repair. In mice, depletion of p75(NTR) exacerbated liver pathology and inhibited hepatocyte proliferation in vivo. In addition, it is showed that neurotrophins activate p75(NTR) to induce apoptosis through the induction of the sphingomyelin (SM) cycle and increased production of ceramide. Overexpression of p75(NTR) is also found to activate the SM pathway.

Principle of the Assay: The Mouse Ngfr Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Mouse Ngfr with a 96-well strip plate that is pre-coated with antibody specific for Ngfr. The detection antibody is a biotinylated antibody specific for Ngfr. The capture antibody is a monoclonal antibody from rat and the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Mouse Ngfr with immunogen: Expression system for standard: NS0; Immunogen sequence: K22-P246. The kit is analytically validated with ready-to-use reagents. To measure Mouse Ngfr, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Mouse Ngfr in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Mouse Ngfr in the sample.

Product Notes

The SCARB1 scarb1 (Catalog #AAA126106) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Scavenging Receptor SR-BI/SCARB1 Antibody Picoband reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Scavenging Receptor SR-BI/SCARB1 can be used in a range of immunoassay formats including, but not limited to, ELISA, FCM/FACS (Flow Cytometry), WB (Western Blot). Researchers should empirically determine the suitability of the SCARB1 scarb1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Scavenging Receptor SR-BI/SCARB1, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
