IF (Immunofluorescence)

(Fluorescent confocal image of Hela cell stained with TARDBP Antibody (N-term). Hela cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with TARDBP primary antibody (1:25, 1 h at 37 degree). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-rabbit antibody (green) was used (1:400, 50 min at 37 degree).Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37 degree). Background: HIV-1, the causative agent of acquired immunodeficiency syndrome (AIDS), contains an RNA genome that produces a chromosomally integrated DNA during the replicative cycle.A ctivation of HIV-1 gene expression by the transactivator Tat is dependent on an RNA regulatory element (TAR) located downstream ofthe transcription initiation site. The protein encoded by this gene is a transcriptional repressor that binds to chromosomally integrated TAR DNA and represses HIV-1 transcription. In addition, this protein regulates alternate splicing of the CFTR gene. A similar pseudogene is present on chromosome 20. [provided byRefSeq].

Function: RNA-binding protein that is involved invarious steps of RNA biogenesis andprocessing (PubMed:23519609).Preferentially binds, via its two RNArecognition motifs RRM1 and RRM2, to GU-repeats on RNA molecules predominantlylocalized within long introns and in the 3'UTRof mRNAs (PubMed:23519609, PubMed:24240615,PubMed:24464995). In turn, regulates the splicing of many non-codingand protein-coding RNAs including proteinsinvolved in neuronal survival, as well asmRNAs that encode proteins relevant forneurodegenerative diseases (PubMed:21358640, PubMed:29438978). Playsa role in maintaining mitochondrialhomeostasis by regulating the processing ofmitochondrial transcripts (PubMed:28794432). Regulatesalso mRNA stability by recruiting CNOT7/CAF1deadenylase on mRNA 3'UTR leading topoly(A) tail deadenylation and thusshortening (PubMed:30520513). Inresponse to oxidative insult, associates withstalled ribosomes localized to stress granules(SGs) and contributes to cell survival(PubMed:23398327, PubMed:19765185).Participates also in the normal skeletalmuscle formation and regeneration, formingcytoplasmic myo-granules and bindingmRNAs that encode sarcomeric proteins(PubMed:30464263).

Product Notes

The TARDBP tardbp (Catalog #AAA284138) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The immunogen sequence is 1-30. The TARDBP Antibody (N-term) reacts with Human, Mouse, Rat; Predicted: Chicken and may cross-react with other species as described in the data sheet. AAA Biotech's TARDBP can be used in a range of immunoassay formats including, but not limited to, ELISA, IHC (Immunohistochemistry), IF (Immunofluorescence), WB (Western Blot). Researchers should empirically determine the suitability of the TARDBP tardbp for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TARDBP, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
